Abstract
Die vorliegende Erfindung betrifft ein Verfahren zur Gewinnung von Bacteriorhodopsin aus Bakterien ueber ein mehrstufiges Reinigungsverfahren.

DE 19945798 C UPAB: 20001109 NOVELTY - Recovery of bacteriorhodopsin (I) from halobacteria (A) comprises growing (A) in nutrient medium, microfiltration to separate (A) and the medium, lysing (A), and microfiltration to eliminate the low molecular weight cell components from (I). The medium contains yeast extract, protein hydrolyzate and/or amino acids or peptides as nitrogen source. USE - (I), a light-driven proton pump in (A), is useful as a light-energy convertor, e.g. for transient optical storage in data processing, in production of biosensors and biochips, or as electro-optical switches, also as macroscopic ion pumps for production of hydrogen. ADVANTAGE - The method is inexpensive, simple (no complicated apparatus) and suitable for scale up. It produces high purity (I) efficiently and can be operated batchwise or (semi)continuously. The use of the specified nitrogen sources (rather than the conventional peptone) increases productivity of (I) and reduces cost.
