Improved Total Synthesis of Cannabioxepane, a Minor Phytocannabinoid from Cannabis sativa L
Abstract
Cannabioxepane, a recently described minor cannabinoid, remains largely unexplored pharmacologically due to its low natural abundance in Cannabis sativa. Previous synthetic approaches have yielded only limited quantities, necessitating the development of an optimized total synthesis. Here, we report a novel synthetic route that circumvents challenges associated with rearomatization by employing prearomatized starting materials. The key carbon–carbon bond formation between the aromatic rings is achieved via palladium-catalyzed C–H activation, providing an efficient strategy for cannabioxepane synthesis.
