Abstract
NOVELTY - Superpotent human calcitonin analogues which exhibit a much increased hypocalcemic activity in vivo are new. DETAILED DESCRIPTION - Calcitonin and calcitonin derivatives comprising a bridge between the amino acids at positions 17 and 21 in the corresponding human calcitonin forming an 18 or 19 membered ring. USE - The calcitonin analogs are useful for lowering calcium levels and especially for treating osteoporosis, Paget's disease and hypercalcemia. ACTIVITY - Hypocalcemic. MECHANISM OF ACTIVITY - Calcium release inhibitor. ADVANTAGE - A 1000 fold increase in activity compared to human calcitonin is observed in in vivo tests in mice.
