Title

Bioreaktor zur Stimulation von einzelnen Zellen, Zellverbuenden und Zellgeweben

Other Title
Bioreactor for stimulation of single cell, cell composite and/or cell tissue, comprises carrier substance, where single cell, cell composite and/or cell tissue are immobilized inside the carrier substance, and temperature control element
Abstract

DE 102009004336 A1 UPAB: 20100806 NOVELTY - The bioreactor comprises a carrier substance (3), where a single cell, cell composite and/or cell tissue are immobilized inside the carrier substance and/or at one of the limiting surfaces of the carrier substance, and a temperature control element, which is arranged, so that the temperature of the carrier substance is controlled using the temperature control element, which is formed as microprocessor-controlled Peltier element (2). The carrier substance has temperature ranges of characteristic temperature point and/or temperature range suited for aqueous and/or aqueous-organic media. DETAILED DESCRIPTION - The bioreactor comprises a carrier substance (3), where a single cell, cell composite and/or cell tissue are immobilized inside the carrier substance and/or at one of the limiting surfaces of the carrier substance, and a temperature control element, which is arranged, so that the temperature of the carrier substance is controlled using the temperature control element, which is formed as microprocessor-controlled Peltier element (2). The carrier substance has temperature ranges of a characteristic temperature point and/or temperature range suited for aqueous and/or aqueous-organic media, where the carrier substance is changeable around the characteristic temperature point by changing the temperature and/or the state of the carrier substance is changeable around the or in the characteristic temperature range. The immobilized single cell, cell composite and/or cell tissue are multi-dimensionally stimulated based on mechanical forces. The temperature of the carrier substance is periodically changeable with a frequency of 0.1-2 Hz. The characteristic temperature point and/or characteristic temperature range is 35-38 degrees C. The hydrogel cyclically includes and transfers water in the frequency range of 0.1-2 Hz. The change of state of the carrier substance that is optionally a gel is a change of swelling states, expansion states, phase states, aggregate states and conformation states and/or a change of chemical properties of the carrier substance. The carrier substance is thermo-reversible i.e. reversible to a temperature change. The carrier substance has reservoirs of endothelial growth factors, fibroblast growth factors, bone-morphogenetic factors and/or tumor necrosis factors and/or active materials and/or concrete materials such as calcium ions (Ca2+-ions) or dexamethasone. The single cell, cell composite and/or cell tissue are vitality-sustainably immobilized in and/or on the biocompatible carrier substance by stimulation in the vicinity of the cell-physiological optimal temperature and/or are vitality-sustainably released from and/or by the carrier substance. An INDEPENDENT CLAIM is included for a method for the stimulation of a single cell, cell composite and/or cell tissue. USE - Bioreactor for the stimulation of single cells, cell composites and/or cell tissues and/or for mechanical stimulation of stem cells and/or cell composites during in-vitro production of supporting tissue and cartilage tissue (claimed). ADVANTAGE - The bioreactor can be economically and simply manufactured, and allows the control of selected cultivation parameters such as carbon dioxide-loading or oxygen-loading, temperature and the media exchange.
