Abstract
(A1) Die Erfindung betrifft einen Protein-Biosensor mit einer definierten Bindematrix, die einen anorganischen oder Kunststoff-Anteil und ein daran angebundenes Protein aufweist.

DE 10006760 A UPAB: 20011217 NOVELTY - Binding matrix (A) comprises: (i) a carrier (I) with a first functional group (G1) on its surface; and (ii) a protein (II), having a functional group (G2) complementary to G1, immobilized on it. DETAILED DESCRIPTION - INDEPENDENT CLAIMS are also included for the following: (1) method for producing (A); (2) method for identifying and/or detecting a binding partner (BP) of (II), using (A); and (3) method for identifying and/or detecting agents that modify (inhibit or promote) interaction between an immobilized (II) and its BP, using (A). USE - (A) Is used, in medical research and diagnosis, to identify and/or detect: (i) binding partners (BP) of (II); or (ii) agents that modulate (promote or inhibit) interaction between BP and (II). Especially it is used to identify BP for the Ras-binding domain (RBD) of Raf-kinase or agents that inhibit interaction of RBD with its BP. (A) can also be used to detect the tumor marker ras-GTP (guanosine triphosphate), which can be differentiated from ras-GDP, e.g. as part of an invasive biosensor. ADVANTAGE - The binding between (I) and (II) is stable (not dependent on external factors (medium, buffer, temperature). (II) can be attached through a sidechain (rather than a terminus) and labeled at any position, so that it retains the native (active) conformation and its ability against chemical or enzymatic decomposition.
