Abstract
Es wird ein Verfahren zur Reinigung von Lipocortinen, besonders der gentechnisch hergestellten Proteine rPP4, rPP4-X und rPP4-delta, mittels Behandlung einer Loesung eines Lipocortins mit einem Anionenaustauscher in Gegenwart eines Chelatbildners und eines Detergen beschrieben.

EP 441274 A UPAB: 19971030 Purificn. of lipocortins (A) comprises treating a lipocortin-contg. soln. with a chelate former and a detergent, contacting the soln. obtd. with an anion exchanger and separating the liq. Also claimed is the further purificn. of (A) by affinity chromatography on an immobilised saccharide polysulphate or on a carrier-bound sulphated sugar or by adsorption on kaolin, Aerosil (RTM) or calcium phosphate. The natural (A) (A) rPP4, rPP4-X and rPP4-delta are described in DE 3910240. A lipocortin-contg. soln. from a yeast, animal cell or E. coli culture is pref. used, as starting material. USE/ADVANTAGE - (A) have anticoagulant and antiinflammatory activity. The method is esp. applicable to the purificn. of rPP4, rPP4-X and rPP4-delta for which no satisfactory purificn. process has hitherto existed.
