Abstract
Die vorliegende Erfindung betrifft ein verbessertes Verfahren zur massenspektrometrischen Analyse, insbesondere zur Matrix-unterstuetztenn Laser-Desorptions/Ionisations-Flugzeit-Massenspektrometrie (MALDI-TOF-MS), unter Verwendung von Nanopartikeln, wobei ein Analyt einer Nanopartikelsuspension zugesetzt wird und die den gebundenen Analyten enthaltende Suspension dann direkt auf einem MALDI-Probentraeger abgeschieden und massenspektrometrisch untersucht wird, sowie fuer dieses Verfahren geeignete Nanopartikel.
WO2003021267 A UPAB: 20030501 NOVELTY - Investigating (M1) at least one analyte (I) by matrix-assisted laser desorption-ionization time-of-flight mass spectrometry (MALDI-TOF-MS) after binding (I) to a nanoparticle, is new. DETAILED DESCRIPTION - M1 includes a first functional group (FG1), to a nanoparticle (NP). NP includes on its surface a functional group (FG2) complementary to (FG1). NP are suspended in an aqueous fluid then at least one (I)-containing sample is added (or vice versa) so that affinity binding links (I) to NP. NP, with attached (I), are deposited on the MALDI sample carrier and analyzed. INDEPENDENT CLAIMS are also included for: (1) NP that carry FG2 on the surface; (2) aqueous suspensions of NP; and (3) a kit comprising NP, as powder or aqueous suspension. USE - M1 is particularly used for analysis of proteins, specifically for BSE tests and for diagnosis of tumors or infectious diseases. ADVANTAGE - Use of NP allows isolation of (I) from complex matrices but NP do not interfere in the MALDI-TOF process, so do not need to be removed, simplifying the analysis.
