Abstract
Die Erfindung betrifft eine Anordnung und ein Verfahren zur optischen Detektion von in Proben enthaltenen chemischen, biochemischen Molekuelen und/oder Partikeln, bei der die sich in Abhaengigkeit vom optischen Brechungsindex veraendernde Oberflaechen-Plasmonen-Resonanz (SPR) fuer die optische Detektion ausgenutzt werden kann. Dabei wird Licht einer Lichtquelle ueber mindestens ein bereichsweise als Lichtwellenleiter ausgebildetes Sensorelement auf einen optischen Detektor gerichtet. Auf einer Grenzflaeche dieses Lichtwellenleiters ist mindestens eine elektrisch leitende Duennschicht unterhalb einer Messflaeche ausgebildet. Das durch den Lichtwellenleiter gefuehrte Licht trifft dabei auf die Grenzflaeche im Bereich der Duennschicht mit Winkeln im Bereich oberhalb des Totalreflexionswinkels auf. Das Sensorelement ist an einer Stirnseite fuer den Lichteintritt so gestaltet, dass eine asphaerisch, konvex gekruemmte optisch wirksame Oberflaeche vorhanden ist, mit der das Licht innerhalb des Sensorelementes in Richtung auf die mit der mindestens einen Duennschicht und/oder Messflaeche belegte Grenzflaeche fokussiert wird. Eine zweite asphaerisch, konvex gekruemmte optisch wirksame Oberflaeche ist an der Stirnseite des Sensorelementes fuer den Lichtaustritt vorhanden. Mittels dieser optisch wirksamen Oberflaeche wird das infolge von Oberflaechen-Plasmonen-Resonanz modifizierte Licht in kollimierter Form auf den mindestens einen optischen Detektor gerichtet.

DETAILED DESCRIPTION - A convex optical focusing surface (6) is shaped at an end side of the sensor (5), which deflects the light and focuses it within the sensor towards at least one thin layer surface (11) and/or a measurement surface (13). A second optical working surface (7), with a convex curvature, is at the end side for the light to pass out, which is modified by the SPR in a collimated form and aligned at the detector (10). An optical diaphragm (4) is between the monochromatic light source (1) and the optical focusing surface. USE - The apparatus is for the detection of analytes in general receptor-ligand systems e.g. with very different antigen-antibody combinations. The biological fluids can contain proteins, molecules, particles and other substances for detection. ADVANTAGE - The system is simple and inexpensive, giving a high detection safety in a wide range of liquid samples without the use of markers.
