Studies on the mode of action of Multi-Walled Carbon Nanotubes onto human bronchial epithelial cells
Abstract
Die Produktion von Nanomaterialien nimmt rasant zu, wobei die jährliche Fertigung von mehreren hundert Tonnen Multi-Walled Carbon Nanotubes (MWCNTs) den größten Teil einnimmt. MWCNTs bestehen aus einzelnen Röhren, die konzentrisch umeinander gelegt sind, und deren Durchmesser weniger als 100 nm beträgt, wobei sie Längen von mehreren Mikrometern erreichen können. Aufgrund dieser an Asbestfasern erinnernden Struktur gibt es starke Bedenken, dass MWCNTs ähnliche Risiken bergen könnten. Da die inhalative Route sicherlich als vornehmlicher Expositionspfad des Menschen für MWCNTs angenommen werden kann, sollte in dieser Arbeit deren Wirkung auf Lungenzellen untersucht werden. Der Einfluss von MWCNTs auf die alveolarepitheliale Zelllinie A549 ist im Rahmen von Toxizitätsscreenings in der Literatur bereits breit diskutiert und wird mit der Induktion von ROS und anderen zytotoxischen Ereignissen in Verbindung gebracht. Im Gegensatz dazu existieren nur wenige Publikationen zur Wirkung von MWCNTs auf Zellen des bronchialen Lungenbereichs, dessen Untersuchung allerdings aufgrund der Depositionsmechanismen von Fasern als relevant anzusehen ist. Zu diesem Zweck wurden in dieser Arbeit in vitro-Zytotoxizitätsassays mit der Bronchialepithelzelllinie Calu-3 durchgeführt. Bei der Bewertung der Zytotoxizitätsassays muss die Interferenz zwischen MWCNTs und olorimetrischen wie auch fluorimetrischen Assaysubstanzen stets berücksichtigt werden. Hierbei konnten lediglich vereinzelt geringe Zellviabilitätseinschränkungen nachgewiesen werden. So wurde nach einer Exposition der Zellen mit 5 ?g/cm² MWCNTs eine Zunahme an Dead-Cell Proteasen im Zellüberstand gemessen und eine Exposition mit 20 ?g/cm² MWCNTs führte zu einer ATP-Abnahme. Durch ihr enges Wachstum in Monoschichten bilden Calu-3-Zellen epithelartige Strukturen, die sich für Untersuchungen zur Stärke des transepithelialen Widerstandes eignen. In dieser Arbeit konnte kein Effekt der MWCNTs auf die Stärke des transepithelialen Widerstandes festgestellt werden. Die Induktion von oxidativem Stress und die Aktivierung des NF?B-Signalweges gelten als wichtig für die Ausbildung faservermittelter Entzündungen. Die Untersuchungen zur Induktion von oxidativem Stress ergaben keinen MWCNT-abhängigen Anstieg von ROS. Im Gegensatz dazu wurden mittels Genexpressionsanalysen einige wenige Gene identifiziert, die an der Regulation von oxidativem Stress beteiligt sein könnten. Neben der Herunterregulierung des Tumorsuppressorgens EGR1 wurde durch die Exposition mit 10 ?g/cm² MWCNTs die Zusammenfassung IX Expression mehrerer Gene verändert, die an der Reparatur von DNA-Schäden oder der Regulation der Immunantwort beteiligt sind. Es lässt sich insgesamt schlussfolgern, dass Calu-3-Zellen, anders als für A549-Zellen beschrieben, relativ unempfindlich auf die Exposition gegenüber MWCNTs reagierten. Für prädiktive Toxizitätsscreenings von MWCNTs ist demnach die Auswahl eines sensitiven Zellmodels essentiell.

With a production rate of several hundred tons per year, Multi-Walled Carbon Nanotubes (MWCNTs) account for the largest part in the increasing production of nanomaterials. Serious concerns have been raised about the safety of MWCNTs due to their asbestos-like structure. As the most likely route of human exposure to MWCNTs is inhalation, the influence of MWCNTs on the alveolar epithelial cell line A549 has already been widely discussed within the scope of toxicity screenings and is associated with the induction of ROS and other cytotoxic events. In contrast, only few publications exist, which discuss the effects of MWCNTs on cells of the bronchial system. Because of MWCNT's mode of deposition, investigation of the impact of MWCNTs in this area is of great importance. Therefore this project presents the results of several in vitro cytotoxicity assays and gene expression analysis carried out with custom made MWCNTs (length 8.57 ?m, diameter 0.085 ?m) on the human bronchial epithelial cell line Calu-3. For separation of agglomerated MWCNTs into single ones, MWCNTs were suspended in Dulbecco's cell culture medium using a sonotrode twice (90% duty cycle, 100% amplitude) for five minutes. Calu-3 cells were incubated for 24 or 48 h with MWCNTs at a concentration of 5, 10, or 20 ?g/cm². When evaluating cytotoxicity assays, the shifting influence of MWCNTs on several absorption spectra should always be considered. Therefore luminescence based cytotoxicity assays were carried out in addition to colorimetric and fluorimetric assays. In regard to reduction of interfering effects, every assay was adapted to the use of MWCNTs by filtering the assay before slight decreases. After an exposure to 5 ?g/cm² MWCNTs, an increase in dead cell proteases in cell supernatants was measured, while an exposure to 20 ?g/cm² MWCNTs led to a decrease in ATP. The induction of oxidative stress and the activation of the NF?B signaling pathway are generally considered to be important for the formation of fiber-mediated inflammation. Our studies on oxidative stress showed no MWCNT-dependent increase of ROS. Pathway-specific real time PCR arrays (Human NF?B Signaling Pathway, Human Oxidative Stress and Antioxidant Defense, Human Stress & Toxicity PathwayFinder, SABioscience/Qiagen) were carried out to determine MWCNT's influence on gene expression levels. Our studies identified the regulation of a few genes which are thought to be involved in the regulation of oxidative stress. Additionally, MWCNTs changed the expression of several genes which are involved in DNA repair or regulation of immune response and down regulated the expression of the tumor suppressor gene EGR1. Due to their tight growth in monolayers, Calu-3 cells form an epithelial-like structure, which can be used for studies on transepithelial resistance. In the present study, no effect of MWCNTs was detected on the strength of the transepithelial resistance. It can be concluded that, unlike A549 cells, Calu-3 cells are relatively insensitive to exposure to MWCNTs. In order to establish a predictive toxicity screening for MWCNTs, the choice of a sensitive cell model is essential.
