Zelllinienentwicklung und Etablierung einer High-Throughput-Methode zur Optimierung einer plattformbasierten Antikörperaufreinigung
Abstract
Monoklonale Antikörper gehören heute zu den bedeutendsten Produkten der pharma-zeutischen Biotechnologie. Ihre Produktion erfolgt in rekombinanten Zelllinien. Gegen Ende des Prozesses muss der produzierte Antikörper aufgereinigt werden. Darum sind die Effizienz der Antikörperaufreinigung sowie das Generieren hochproduzierender Zellen mit einer vorhersagbaren Antikörperexpression für Industrie und Forschung wichtig. In dieser Arbeit wurden CHO-K1-Zellen im Rahmen eines Rekombinase-vermittelten Kassettenaustauschs (RMCE) mit vier antikörperkodierenden Plasmiden transfiziert. Die spezifischen Antikörperbildungsraten der Zellen wurden ermittelt und verglichen. Ziel der Arbeit war festzustellen, ob die Expressionsraten der verschiede-nen Antikörpergene auf einem gleichen genomischen Locus ähnlich sind. Es konnte belegt werden, dass der RMCE erfolgreich war. Allerdings waren die ermittelten spezi-fischen Produktbildungsraten zu gering, um einen Vergleich durchzuführen. Außerdem wurden CHO-DHFR-DG44-Zellen des Fraunhofer-Institutes auf konventio-nelle Weise mit einem GFP-Gen-tragenden Plasmid transfiziert. Das Ziel war ein Proof of Principle durchzuführen, der belegt, dass die verwendete Zelllinie erfolgreich mit einem Plasmid transfiziert werden kann. Mittels der FACS-Methode konnten hochpro-duzierende Zellklone isoliert und anschließend kultiviert werden. Ein Zellklon konnte über mehrere Wochen erfolgreich kultiviert werden. Des Weiteren wurde die zweistufige chromatographische Aufarbeitung eines monoklo-nalen Antikörpers über eine MabSelect SuRe- und eine Capto adhere-Säule optimiert. Dazu wurden 96-Well PreDictor-Platten in einem High-Throughput-Prozess (HTPD) verwendet und die Ergebnisse einer statistischen Versuchsplanung ausgewertet. Die Ergebnisse wurden auf Chromatographiesäulen übertragen. Daraus resultierte eine Prozessausbeute von 76 %, ein spezifischer HCP-Gehalt von 12 pg/mg sowie ein DNA-Gehalt von 438 pg/mg Antikörper. Zudem konnte die Effizienz des HTPD in Kom-bination mit der statistischen Versuchsplanung festgestellt werden.

Nowadays monoclonal antibodies are one of the most important products in pharma-ceutical biotechnology. They are produced in recombinant cell lines. Finally, a pro-duced antibody has to be purified to eliminate impurities like HCP or DNA. Therefore, efficiency of purification and generation of high producing cells is a key factor in the production of antibodies in science and industry. Additionally, the possibility of estimat-ing the antibody production of a cell would be high of value. In this work CHO-K1 cells were transfected with four different vectors containing gens for expressing a human IgG 1, IgG 2, IgG 4 or a murine IgG 2a. The productivity of the cells was compared with respect to the production of the different IgGs. The objective of this work was to deter-mine if the productivity of the cells is comparable for the four antibodies which were integrated into the same locus. Results indicated that the RMCE event was successful. However, the production of antibodies was too low to compare the productivity of the cells with respect to the four different classes of antibodies. Furthermore, CHO-DHFR-DG44 cells of the Fraunhofer-Institute were transfected with a plasmid which encodes for GFP. The GFP encoding gen was integrated into the ge-nome of the cell randomly. The objective was to conduct a proof of principle that indi-cates if the cell line could be transfected with a gen of interest successfully. By sorting cells in a FACS, it was possible to isolate and to cultivate high producing cells. Finally, a low producing cell was cultivated over several weeks. In addition the purification of a monoclonal antibody was optimized in a two step purifi-cation process. The purification contained a capture step on MabSelect SuRe and a polishing step on Capto adhere. 96 Well PreDictor Plates were used in a high through-put process development (HTPD) to evaluate the best purification conditions. Experi-ments were based on central composite Design of Experiments. The results from the PreDictor Plates were transferred to columns. Thereby, the purification strategy caused a yield of 76 %, a HCP content of 12 pg/mg and a DNA content of 438pg/mg. Finally, the efficiency of HTPD and Design of Experiments could be assessed.
