Abstract
(A1) Die Erfindung betrifft ein Verfahren zum Detektieren einer oder mehrerer chemischeer oder biologischer Spezies, die entweder eine Redox-Reaktion eingehen koennen oder direkt oder indirekt ein Molekuel generieren koennen, das eine Redox-Reaktion eingehen kann, wobei durch die genannte Redox-Reaktion generierter Strom an mindestens einer Elektrode detektiert wird, umfassend die folgenden Schritte: 1. Positionieren einer zeitlich variierenden Menge der Spezies oder des Molekuels auf, an oder in der Naehe der mindestens einen Elektrode innerhalb eines Zeitraum t?1?-t?2?, 2. mehrfaches Hin- und Herschalten der mindestens einen Elektrode waehrend des Zeitraums t?1?-t?2? zwischen zwei unterschiedlichen Potentialen, derart, dass sie gegenueber einer Referenzelektrode Potentiale einnimmt, die im Bereich des Oxidationspotentials der genannten Spezies oder des genannten Molekuels oder darueber bzw. im Bereich des Reduktionspotentials der genannten Spezies oder des genannten Molekuels oder darunter liegen, wodurch die genannten Spezies/das genannte Molekuel abwechselnd reduziert und oxidiert wird, und 3. Erfassen des ueber den Zeitraum 7?1?-t?2? durch wiederholte Reduktion und Oxidation der Spezies/des Molekuels in der mindestens einen Eltkrode gebildeten Stroms. DETAILED DESCRIPTION - Detecting one or more chemical or biological species, which either undergo a redox reaction or directly or indirectly generate a molecule undergoing a redox reaction, comprises detecting the current generated by the redox reaction at an electrode by the following steps: positioning temporally varying amount of the species or molecule, or in the vicinity of the electrode within a time period of t1-t2; repeated switching of the electrode during the time period of t1-t2 between two different potentials such that it assumes potentials with respect to a reference electrode lie within the range of the oxidation potential of the species or molecule, or above or within the range of the reduction potential of the species or molecule, or lie below it, where the species/molecules are alternately reduced and oxidized; and detecting the current which is formed in the electrode over the time period of t1-t2 by repeated reduction and oxidation of the species/molecule. INDEPENDENT CLAIMS are included for: (1) an electrode array for carrying out the above method, comprising at least a measurement position, at least a measurement electrode per measurement position and a reference electrode, where the electrode array is designed in such a way that the measurement electrode is switched with respect to the reference electrode alternately as cathode and anode; and (2) an apparatus for carrying out the above method, comprising the electrode array, a counter electrode, a reference electrode and a circuit arrangement. USE - The method is useful for detecting chemical or biological species such as proteins, preferably enzymes e.g. phosphatases (claimed), molecular complexes, peptides, antigens, nucleic acids and nucleic acid containing species. ADVANTAGE - The method avoids short circuits and inter-digital electrodes. The electrodes used in the method are easy to made and less expensive.
