Untersuchungen zum Einfluß der Proteintyrosin-Phosphatase SHP-1 auf den CSF-1 vermittelten Signaltransduktionsweg in Makrophagen
Abstract
The CSF-1 induced signalling pathway in macrophages will be initiated through the binding of CSF-1 to the CSF-1 receptor. After CSF-1 binding the CSF receptor will be autophosphorylated. Immediately both the serine/threonine kinase RAF-1, the extracellular and mitogen activated kinase MEK and the mitogen activated protein kinase MAPK will be phosphorylated. Subsequently the activated MAPK translocates into the nucleus leading to the activation of specific transcription factors and thereby gene expression. The antagonistic operating phosphatases deactivate kinases by dephosphorylation. One of these phosphatases recently well investigated is the protein tyrosine phosphatase SHP-1 which is predominantly expressed in hematopoietic cells. As SHP-1 is involved into the CSF-1 signalling pathway the enzyme seemed to be very important for macrophages growth, differentiation and survival.
