Abstract
Sphaerische magnetische Silica-Partikel mit einer Teilchengroesse von 0,5-2000 ?m und einstellbarer Porengroesse werden mit Hilfe eines Wasser-in-Oel-Dispersions-Vernetzungsverfahrens von Silica-Solen hergestellt. Durch Mischen der Silica-Sole mit organischen Polymeren lassen sich Silica-Partikel mit veraenderten physikalischen und chemischen Eigenschaften herstellen. Die Magnetpartikel lassen sich zur Nukleinsaeure-Separation sowie zur Isolierung von Biomolekuelen einsetzen.
DE 10035953 A UPAB: 20020521 NOVELTY - Preparation of spherical, magnetic silica particles (A) by; (i) mixing a silica hydrosol (B) with a magnetic colloid or ferrofluid, or with magnetic particles; (ii) dispersing the mixture in a water-immiscible organic solvent phase and (iii) during dispersion, crosslinking by adding base. (B) is produced by acid-catalyzed hydrolysis of an aqueous silane dispersion. DETAILED DESCRIPTION - An INDEPENDENT CLAIM is also included for (A) of particle size 0.5-2000 micron, in which the pore size and content of magnetic component can be adjusted. USE - (A) are used for isolation and analysis of optionally biotinylated nucleic acid, proteins and antibodies (or their fragments). ADVANTAGE - (A) are prepared without using laborious or lengthy coating techniques, and the pore and particle sizes, and content of magnetic component, can be adjusted. They provide efficient separation of biomaterials and nucleic acid, without the problems of polymerases being inactivated by contact with iron compounds. Surfaces of (A) can be modified either to allow attachment of biomolecules, or by polymers to alter adsorption properties for proteins, mechanical properties or stability towards bases.
