Effects of polychlorinated dibenzo-p-dioxins in epidermal metabolism of euthyroid and hypothyroid C57BL/6J mice
Abstract
Female C57BL/6J mice at the age of seven weeks were functionally thyroidectomized with 131I (7,5 mCi, i.p.). Thereafter, a iod deficient diet was fed. A single oral administration of tetrachlorodibenzo-p-dioxin (TCDD) as well as mixtures of polychlorinated dibenzo-p-dioxins (PCDD) was followed 10 weeks later. At the time of treatment, the hypothyroid status was demonstrated by a lowered serum concentration of thyroxine and an enhanced cholesterol concentration. Simultaneously, TCDD and PCDD were administered also to euthyroid C57BL/6J mice, 4 and 7 days after TCDD treatment, ear skin of six hypothyroid and euthyroid mice was sampled. The incorporation rates of thymidine triphosphate into DNA and of leucine and histidine into protein as well as the ornithine decarboxylase activity were measured in epidermis separated from the skin samples.
