Abstract
The invention relates to a process for detecting alkaline phosphatase in cells by incubating the cells with an alcaline solution of an aromatic phosphite and a diazonium salt, and converting the aromatic alcohol formed by the alkaline phosphatase from the phosphite via an azo coupling into an azo compound, and azo dye by means of the diazonium salt, the azo dye being continuously precipitated from the diaphragm. This azo dye is selectively eluted from the cell membrane by means of a suitable organic solvent and is quantitatively detected by measuring the extinction of the resulting organic solution at a suitable wavelength compared with a corresponding blank reading. The volume and the activity of the alkaline phosphatase is quantitatively determined from the concentration and from the volume of the azo dye formed and from the number of the analyzed cells. The preferred phosphites are phosphites from derivatives of the 3-hydroxy-2-naphthol AS such as naphthol AS-MX, naphthol AS-BI and naphthol AS-TR. The preferred diazonium salts are Fast Red TR, Fast Red Violet LB, Fast Blue BB or Fast Blue RR.
