Nasal retention and clearance of wood dust in rats

The objective of this study was to develop an animal model in which the nasal cavities can be tested as a target organ for wood dust. The intranasal instillation and the inhalation route were investigated as alternative treatment methods. A wood dust fraction with a mass median aerodynamic diameter of 4-5um was used which was labelled by the 51Cr tracer technique. An apparent deposition fraction of about 25% was determined by inhalation. The clearance halftimes were 200-250 min for both administration methods. However, the standard deviation was higher after intranasal installation. The reproducibility of dosing improved after inhalation exposure which leads to a more even distribution of particles in the nasal cavities. Histopathological investigations showed inflammatory processes after repeated intranasal instillation. (Abstract truncated)
