Characterisation of an automated bioreactor system for osteogenic differentiation of murine embryonic stem cells
Abstract
Seit einiger Zeit rücken pluripotente embryonale Stammzellen (ESCs) zunehmend in den Fokus der Wissenschaft. Sie werden vielseitig in der regenerativen Medizin und für Entwicklungstoxizitätstests eingesetzt. Die Differenzierung embryonaler Stammzellen ist für die statische Kultur gut etabliert. Die vorliegende Arbeit stellt ein automatisiertes Bioreaktorsystem vor, mit dessen Hilfe Embryoid Bodies (EBs) aus murinen ESCs erzeugt und zu Osteoblasten differenziert wurden. Bioreaktoren bieten den Vorteil Prozessbedingungen wie Temperatur, pH-Wert, sowie CO2- und O2-Gehalte exakt zu regeln. Darüber hinaus sorgt die gleichmäßige Durchmischung für eine verbesserte Nährstoffversorgung. Im Verlauf der Arbeit wird die spezifische Wachstumsrate µ für Maus-ESC bestimmt. Des Weiteren erfolgte eine Einschätzung der Zelldifferenzierung in Abhängigkeit von dem verwendeten Rührertypen und der genutzten Drehzahl. Dazu wurden Expressionsanalysen der Pluripotenzgene Oct4 und nanog, sowie spezifischer Keimbahngene durchgeführt. Eine Analyse der EB-Formierug erfolgte anhand einer EB-Größenverteilung und Größenvergleichsmessungen. Die Differenzierung zu Osteoblasten wurde erfolgreich mit EBs aus einer hängenden Tropfenkultur und mit im Bioreaktor gebildeten EBs durchgeführt.

In the last years pluripotent embryonic stem cells (ESCs) have increasingly been focused in science. ESCs are used for tissue engineering in regenerative medicine or for embryotoxicity tests. The differentiation of ESCs is well established in static culture. The following task presents the potential of automated bioreactors, to form embryoid bodies (EBs) of murine ESCs and the differentiation of the cells to osteoblasts. Process parameters like temperature, pH, CO2- und O2-concentration can be easily controlled in bioreactors. Additionally, the optimised homogenisation enhances the nutrient supply. Moreover, the growth rate µ of murine ESCs will be determined. Furthermore, the influence of the stirrer typ and the agitation rate to the cell differentiation is examined. To check the cellular development, gen expression markers such as Oct4 and nanog for pluripotency and germ layer markers were analysed. The EB formation was evaluated by comparing EB size and observing of the EB size distribution. A differentiation to osteoblasts was successfully done with cells from hanging drop culture and with cells from the bioreactor.
