Abstract
Verfahren zur Immobilisierung von Molekuelen an Oberflaechen mittels weicher Nanopartikel fuer die Herstellung von Multiparameter-Immuntests. Nachteilig an den bekannten bioanalytischen Immuntests ist die unspezifische Adsorption von Proteinen, das Erfordernis der Aktivierung der reaktiven Gruppen sowie die mangelnde Langzeitstabilitaet. Das erfindungsgemaesse Verfahren loest diese Probleme durch die Einfuehrung von "weichen" Nano- und Mikropartikeln mit einer grossen Zahl von koppelbaren Gruppen als Hilfsstoffen fuer die Kopplung. Das Verfahren ist besonders dazu geeignet, preiswerte miniaturisierte bioanalytische Testsysteme (z. B. Microarrays) auf reaktionstraegen Oberflaechen wie passivierten Glas- oder Polymeroberflaechen mit geringer Oberflaechenenergie herzustellen.

(I) is immobilized on the surface of a solid material by direct adsorption. DETAILED DESCRIPTION - An INDEPENDENT CLAIM is also included for the preparation of a carrier as above. USE - The carrier is useful in detection procedures, in parallel binding assays and in competitive immunoassays (all claimed). The method is useful for immobilization of low molecular ligands (e.g. active agent candidates, metabolites and toxins, including steroid hormones, creatinine, anesthetics and/or pesticides) and high molecular ligands (e.g. antibodies, antigens and peptides) on surfaces of solids. The method is particularly useful in the production of microarrays or biochips. ADVANTAGE - (I) allows the ligands to be irreversibly immobilized on the solid support with high density. (I) reduces non-specific adsorption of proteins. The method produces a biocompatible surface with minimal non-specific binding. The need to take measures to reduce undesired adsorption effects is thus eliminated which reduces production costs and increases product quality. The increase in effective surface area due to the use of nanoparticles increases achievable physical, chemical and biochemical signals.
