Herstellung und Charakterisierung von rekombinanten scFv-Fusionsproteinen zur Generierung Aspergillus flavus-resistenter Maispflanzen
Abstract
Das Ziel dieser Arbeit war die Herstellung von Aspergillus-spezifischen monoklonalen Antikörper (mAks) und korrespondierenden scFv-Antikörperfragmente, welche fusioniert mit antifungalen Peptiden (AFP) als AFP-scFv Fusionskonstrukte in Maispflanzen exprimiert werden sollten, um zu einer Erhöhung der Resistenz gegen aflatoxigene Pilze in Zea mays zu führen. Zur Herstellung von Aspergillus-spezifischen mAks in Hybridomazelllinien wurden Mäuse mit Zellwandfragmenten von A. flavus und A. parasiticus immunisiert und mittels ELISA, Immunoblot und mittels Immunofluoreszenzmikroskopie charakterisiert. Dabei wiesen die Antikörper mAkAP3, mAkAF1 und mAkAP10 die höchsten Reaktivitäten gegen A. flavus und A. parasiticus auf, wobei eine Reaktivität ausschließlich spezifisch gegen Aspergillus spp. und nicht gegen weitere pilzliche Pathogene bestimmt werden konnte. Nach der Identifizierung der Antikörper-kodierenden cDNA-Sequenzen der drei mAks konnte die Funktionalität der in N. tabacum produzierten korrespondierenden scFv-Antikörperfragmente bestätigt werden. Das Antigen des mAkAP10, das Cystein-reiche sekretierte Protein (CSP; XP_002374495.1) konnte erstmalig von einem zuvor generierten Aspergillus-spezifischen Antikörpers mittels Immunoaffinitätschromatographie isoliert und die Funktion von CSP-spezifischer siRNA auf A. flavus und A. parasiticus untersucht werden. Durch vollständige Inhibierung der pilzlichen Keimung für beide Pathogene bei 0,5-1 nM CSP-spezifischer siRNA konnte dabei ein geeignetes Target zur Herstellung von Aflatoxin-resistenten Pflanzen identifiziert werden.Bei der Selektion von Aspergillus-inhibierenden Komponenten konnte erstmals eine starke Reduzierung der Keimung von A. flavus (MIC 4 µM) und A. parasiticus (MIC 12,5 µM) durch Thanatin ermittelt werden. Da die Bindung von rekombinanten Thanatin-scFv und Chitinase-scFv Fusionsproteinen an Aspergillus bestätigt werden konnte, wurden die Thanatin- bzw. Chitinase-scFv Fusionskonstrukte, sowie entsprechende Kontrollkonstrukte zur stabilen Transformation von Mais verwendet. Nach der Transformation von Mais konnte für alle verwendeten Konstrukte die Detektion der integrierten DNA-Konstrukte in transgenen Maispflanzen mittels PCR erfolgen. Zur Verifizierung der Resistenz von transgenen Mais gegen A. flavus wurde das Saatgut im Kernel Screening Assay getestet. Durch die alleinige Expression des Aspergillus-spezifischen scFvAP3 konnte keine erhöhte Resistenz gegen A. flavus nachgewiesen werden. Hingegen konnte im Vergleich zu Wildtypsaatgut, eine bis zu dreifach höhere Toleranz gegen A. flavus, sowohl bei alleiniger Expression von Thanatin und der Chitinase als auch in Fusion mit einem scFv erreicht werden.

The aim of the thesis was to generate Aspergillus-specific monoclonal antibodies (mAbs) and corresponding scFv-antibody fragments, while the recombinant scFv antibodies should be fused to antifungal peptides (AFPs). Maize plants transformed with AFP-scFv fusion constructs should posses an increased resistance against aflatoxigenic fungi in Zea mays. Mice were immunized with cell wall fragments of A. flavus and A. parasiticus to generate hybridoma cell lines secreting Aspergillus-specific mAbs, which were characterized by ELISA, immunoblot and immunofluorescence microscopy. Highest reactivity against A. flavus und A. parasiticus was obtained by mAbAP3, mAbAF1 and mAbAP10, while these mAbs were not reactive to other tested fungi except Aspergillus spp. The antibody-encoding cDNA sequences of the three mAbs were identified and functional recombinant scFvs were produced in N. tabacum. The antigen of mAbAP10, the cysteine-rich secreted protein (CSP; XP_002374495.1) was isolated by immunoaffinity chromatography and the effect of CSP-specific siRNA was analyzed against A. flavus and A. parasiticus. Full inhibition of fungal germination was observed for both pathogens at 0.5-1 nM CSP-specific siRNA. Therefore CSP could be used as a target to generate Aspergillus and aflatoxin-resistant crop plants.For the first time, a strong reduction of fungal germination could be demonstrated for A. flavus (MIC 4 µM) und A. parasiticus (MIC 12.5 µM) using thanatin. Since the functionality of transient-produced thanatin- and chitinase-scFv fusion proteins could be confirmed, these fusion constructs and appropriate control constructs were used for stable transformation in maize. After transformation the integration of all constructs into the genome of maize could be confirmed by PCR. To analyze the resistance of transgenic maize, obtained seeds were tested in kernel screening assay against A. flavus. Compared to inoculated wild type seeds an exclusive expression of Aspergillus-specific scFvAP3 did not lead to an increased resistance against A. flavus. However, a three-fold higher resistance against A. flavus could be detected for transgenic seeds expressing both chitinase and thanatin as well as the chitinase- and thanatin-scFv fusion constructs.
