Purification and characterization of the plant produced human surfactant protein D (hSP-D)
Abstract
Menschliches SP-D ist ein Mitglied der Collectin-Familie und war ursprünglich in der Lunge identifiziert worden wegen seiner Rolle bei durch oberflächenaktive Substanzen vermittelter angeborener Immunität. Sein Mangel kann ernsthafte Krankheiten wie RDS und Asthma auslösen. Heutzutage werden kommerzielle oberflächenaktive Mittel während Behandlungen von Lungenkrankheiten verabreicht, jedoch enthalten diese Mittel kein hSP-D, da dieses hydrophile Protein während der Extraktion von oberflächenaktiven Mitteln aus Tierlungen verloren geht. Heterologe Expressionsplattformen wie Säugetier-Zellen und Bakterien warden zur hSP-D-Produktion verwendet. Jedoch ist die Ausbeute bei Verwendung von Säugetierzellen gering und die Produktionskosten sind hoch und Bakterien besitzen nicht die benötigte Fähigkeit posttranslationale Modifikationen durchzuführen. Pflanzen stellen ein alternatives System zur Produktion von rekombinanten Proteinen dar. Sie erlauben die Produktion im Großmaßstab mit einem hohen Sicherheitsprofil, da sie das Wachstum von menschlichen Erregern nicht unterstützen. Rekombinante Proteine können zu höheren Mengen akkumuliert warden, wenn sie in intrazellulären Kompartimenten ausgesetzt warden. Dies hat Auswirkungen auf post-translationale Modifikationen und auf die Ausbeute. Die Arbeit dieser Thesis fokussiert sich auf die Produktion, Purifikation und Charakterisierung von drei rhSP-D Varianten (hSP-D in voller Größe, eine verkürzte Form hNCRD und einem hNCRD-DsRed fusion-Protein) in Tabak Pflanzen.Die rhSP-D Varianten werden im Apoplast und Cytosol expressioniert um die besten Produktionsbedingungen für die funktionalen Proteine zu finden.Der Apoplast war in Bezug auf die rhSP-D Ausbeute besser geeignet als das Cytosol (30-180 mg/kg FLW). Transgene Tabakpflanzen, welche die Apoplast-gerichteten Proteine expressionierten, zeigten eine vielversprechende Eignung als nachhaltige Produktionsplattform, da diese (60-80 mg/kg FLW) im Vergleich zu Säugetier-Zellen (~2-5 mg/l) und Bakterien (~40 mg/l) höhere Ausbeuten der rhSP-D Varianten ermöglichen. Jedoch ergaben sich Herausforderungen bei der Purifikation von unmarkierten und His6-markierten rhSP-D Varianten, welche aus transienter Expression hervorgingen. Daran beteiligt waren: Starke Bindungen der rhSP-D Varianten an das Chromatographie-Harz, was die Elution hemmte und die Anwesenheit von aus den Pflanzen stammenden Verunreinigungen, welche das Faulen von Chromatographie-Säulen verursachte und mit den Bindungen interferierte. Dies zusammen sorgte letztlich für eine relativ niedrige Ausbeute nach der Purifikation: ~24 mg/l für unmarkiertes hSP-D in voller Größe, ~15 mg/l für unmarkiertes hNCRD, ~16 mg/l für hSP-D in voller Größe mit N-terminal His6 Markierung, ~27 mg/l für hNCRD mit N-terminal His6 Markierung, ~2 mg/l für hSP-D in voller Größe mit C-terminal His6 Markierung und ~10 mg/l für hNCRD mit C terminal His6 Markierung.Competitive ELISA bestätigte die lectinbindende Aktivität der unmarkierten und N-terminal His6-markierten hSP-D in voller Größe und hNCRD Varianten mit den gleichen Saccharidpräferenzen, wie dem nativen hSP-D (N-acetylmannosamin gefolgt von Maltose, Glukose und Galaktose). Die rhSP-D Varianten mit C-terminal His6 Markierungen bindeten nicht an mannanbeschichteten Tellern in den Competitive ELISA Experimenten, was darauf hindeutet, dass die C-terminal Markierung das CRD blockierte und Ligandenbindung dadurch verhindert wurde. Obwohl nur teilweise purifizierte rhSP-D Varianten erhalten wurden, demonstrierte der Bakterien- Agglutinationstest die Kapazität von unmarkiertem hSP-D in voller Größe. Ebenfalls wurde demonstriert, wie das hNCRD-DsRed Fusionsprotein bakterielle LPS bindete und Agglutination induziert wurde. Dennoch waren die Absorptionsraten dieser beiden rhSP-D Varianten etwa zwei mal niedriger als beim hSP-D in voller Größe aus seiner Murine Myeloma Zelllinie. Ihre Aktivität ging nach zwei Monaten Lagerung bei drei verschiedenen Temperaturen verloren. Daher sind weitere Studien notwendig, um ein effizientes und ökonomisches Purifikationsverfahren im Großmaßstab zu entwickeln, welches die effiziente Rückgewinnung von reinem, aus Pflanzen gewonnenem rhSP-D ermöglicht und dabei die biologische Aktivität des Proteins erhält.

Human SP-D is a member of the collectins family and was initially identified in the lung due to its role in surfactant-mediated innate immunity. Its deficiency can cause serious diseases, such as RDS and asthma. Nowadays, commercial surfactants are administered during the treatment of pulmonary diseases, but these products do not contain hSP-D because this hydrophilic protein is lost during the extraction of surfactant from animal lungs. Heterologous expression platforms such as mammalian cells and bacteria are used for the production of hSP-D, but the yields are low and the cost of production is high when using mammalian cells, and bacteria lack the ability to carry out post-translational modifications. Plants offer an alternative system for the production of recombinant proteins, allowing large-scale production and a high safety profile because they do not support the growth of human pathogens. Recombinant proteins may accumulate to higher levels when secreted or directed to intracellular compartments, thus affecting post-translational modifications and yields. The work described in this thesis focused on the production, purification and characterization of three rhSP-D variants (full-size hSP-D, hNCRD, and an hNCRD-DsRed fusion protein) in tobacco plants. The rhSP-D variants were expressed in the apoplast and cytosol to find the best conditions for their production as functional proteins. The apoplast was better than the cytosol in terms of rhSP-D yields (30-180 mg/kg FLW) and transgenic tobacco plants expressing the apoplast-targeted proteins showed promise as a sustainable production platform offering higher yields of the rhSP-D variants (60-80 mg/kg FLW) compared to mammalian cells (~2-5 mg/l) and bacteria (~40 mg/l). However, purification of the untagged and His6-tagged rhSP-D variants following transient expression revealed challenges involving strong binding of the rhSP-D variants to the chromatography resin, which inhibited elution, and the presence of plant-derived contaminants, which caused column fouling and interefered with binding, together resulting in relatively low yields after purification: ~24 mg/l for untagged full-size hSP-D, ~15 mg/l for untagged hNCRD, ~16 mg/l for full-size hSP-D with an N-terminal His6 tag, ~27 mg/l for hNCRD with an N-terminal His6 tag, ~2 mg/l for full-size hSP-D with a C-terminal His6 tag and ~10 mg/l for hNCRD with a C terminal His6 tag. Competitive ELISA confirmed the lectin-binding activity of the untagged and N-terminal His6-tagged full-size hSP-D and hNCRD variants with the same saccharide preferences as the native hSP-D (N-acetylmannosamine followed by maltose, glucose and galactose). The rhSP-D variants with C-terminal His6 tags failed to bind mannan-coated plates in the competitive ELISA experiments, suggesting the C-terminal tag blocked the CRD and prevented ligand binding.Although only partially-purified rhSP-D variants were obtained, the bacterial agglutination assay demonstrated the capacity of the untagged full-size hSP-D and the hNCRD-DsRed fusion protein to bind bacterial LPS and induce agglutination. Nevertheless, the absorbance values of these two rhSP-D variants were ~2-fold lower than the full-size hSP-D control from a murine myeloma cell line, and their activity was complete lost after two months in storage at three different temperatures. Further studies are therefore required to develop an efficient and economical large-scale purification method that can achieve the efficient recovery of pure plant-derived rhSP-D and maintain the biological activity of the protein.
