Abstract
Based on the previously developed microfluidic library for SimulationX a ""model-in-the-loop"" example for a branched microfluidic cell culture system is presented. These in vitro drug screening platforms are getting more and more important for pharmaceutic and cosmetic substance evaluation and may replace animal testing in the next years. By exporting the validated model of the microfluidic system to the pneumatic controlling unit (BeagleBone black embedded linux system) which actuates the pumps and valves on the chip one can regulate nutrient supply to the cell culture segments. This is from special interest for opaque high-density tissue cultures where optical sensors cannot be used. With the presented approach one can count back from a reference channel to ensure sufficient nutrient supply and in vivo like cultivation conditions in the black box cell culture chamber. The proof of principle is carried out by operating the chip-integrated micro pump with different process gases (nitrogen, compressed air) in a branched microfluidic system with two parallel cell culture segments. By actuating the upstream valves a certain oxygen level in each branch can be established.

Auf Basis der entwickelten mikrofluidischen Bibliothek für SimulationX soll ein komplexes, verzweigtes mikrofluidisches System für die Zellkultivierung ausgeregelt werden. Solche sogenannten ""in-vitro-Tox-Tests"" werden zunehmend als Ersatz für Tierversuche in der pharmazeutischen und kosmetischen Wirkstofftestung eingesetzt. Mit dem vorgestellten ""model-in-the-loop""-Ansatz lassen sich Sensoren für das Online-Monitoring einsparen und gleichzeitig in-vivo-ähnliche Bedingungen für die Zellkulturen nachbilden. Dafür soll ein validiertes Modell des mikrofluidischen Systems entwickelt und in die zur Regelung der Aktoren (Ventile, Pumpen) verwendete pneumatische Steuerung auf Basis eines BeagleBone Black exportiert werdem. Die Steuerung berechnet dann anhand des Modells wie die Stellglieder zu setzen sind, um zum Beispiel entsprechend des Verbrauchs die einzelnen Kompartimente mit Nährstoffen zu versorgen. Ein Sonderfall stellen dabei intransparente Gewebekulturen dar, in welchen eine optische Messung nicht möglich ist. Um in dieser Blackbox eine ausreichende Nährstoffversorgung zu gewährleisten muss mit dem Modell von einem, mit dem Messsystem erreichbaren, Referenzkanal auf die Kulturkammer zurückgerechnet werden. Das Wirkprinzip wird anhand der gezielten De- bzw. Oxygenierung in einem mikrofluidischen System mit zwei parallelgeschalteten Zellkulturbereichen gezeigt.
