Surrogate development and characterization for the performance check of ApoStreamTM
Abstract
The aim of the present thesis was to develop and characterize surrogates for the ApoStreamTM performance check which mimic both circulating tumor cells (CTCs) and peripheral blood mononuclear cells (PBMCs) with respect to their behavior in an inhomogeneous electric field. Non-biological and biological particles should be developed which undergo positive (like CTCs) or negative dielectrophoretic (DEP) force (like PBMCs) with a minimum shelf life of six months. Therefore, modified PS-beads and different fixed cells were first investigated for their DEP behavior in a stationary inhomogeneous electric field with the ApoCell Analyzer. Furthermore, the stability over time periods and different storage conditions were tested to determine possible changes in behavior. Based on these results, the performance of the most promising surrogates was checked in the ApoStreamTM. In addition, ApoStreamTM settings were changed to optimize the surrogate performance in the device. Pegylated PS-beads are good candidates to represent PBMCs in the ApoStreamTM due to the prevention of sticking resulting from the PEG and their density close to the normal blood cells as well as their negative DEP behavior. Paraformaldehyde fixed, glycine blocked cells from the primary cancer cell line MT_007 could mimic CTCs in this device, because they have a positive DEP behavior and their size is comparable to CTCs. A preliminary ApoStreamTM run showed that further investigations are necessary to optimize the performance of the fixed cells. Consequently, surrogates were developed that basically fulfill the requirements, but further investigations are necessary to optimize the performance of the fixed MT_007 cells. Pegylated PS-Beads can be used as surrogates for PBMCs.

Das Ziel der vorliegenden Diplomarbeit war die Entwicklung und Charakterisierung eines Standards für den Funktionsnachweis des ApoStreamTM. Das Verhalten der zu entwickelnden biologischen und nichtbiologischen Surrogate sollte dem der zirkulierender Tumorzellen (CTCs) und der mononuklearer Zellen des peripheren Blutes (PBMCs) in einem inhomogenen, elektrischen Feld entsprechen, wobei CTCs eine positive und PBMCs eine negative dielektrophoretische (DEP) Kraft erfahren. Die Surrogate sollten eine Lagerstabilität von mindestens sechs Monaten aufweisen. Die Auswahl erfolgsversprechender Surrogate zur weiteren Untersuchung im ApoStreamTM erfolgte durch die Ermittlung des DEP Verhaltens modifizierter PSBeads und verschiedener, fixierter Zellen in einem stationären, inhomogenen, elektrischen Feld mittels dem ApoCell Analyzer. Durch Variation der ApoStreamTM-Einstellung konnte das Verhalten der Surrogate im Gerät optimiert werden. Des Weiteren wurden die Langzeit-Stabilität sowie der Einfluss verschiedener Lagerungsbedingungen auf mögliche Veränderungen im Partikelverhalten bestimmt. Pegylierte PS-Beads erwiesen sich als gute Standardpartikel für PBMCs, da sie in ihrer Dichte normalen Blutzellen ähneln und wie diese einen negativen DEP Effekt besitzen. Paraformaldehyd-fixierte, Glycin-geblockte Zellen der primären Krebszelllinie MT_007 eigneten sich als Standardpartikel für CTCs, da sie ein positives DEP Verhalten aufweisen und in ihrer Größe den nachzuweisenden CTCs entsprechen. Erste Messungen zeigten jedoch, dass die Wiederfindungsrate relativ gering ist. Somit konnten Standardpartikel entwickelt werden, mit denen grundsätzlich das gewünschte Ergebnis erzielt wird. Es besteht jedoch ein Optimierungspotential hinsichtlich der Wiederfindungsrate der fixierten MT_007. Die pegylierten PS-Beads können als Standard für die PBMCs verwendet werden.
