Influence of recombinant Wnt proteins on osteoblast differentiation of murine embryonic stem cells in serum free mediumSta
Abstract
Bone is a mineralized tissue that comprises different mechanical and metabolic functions, which are then carried out in the skeleton. Bone is composed of distinct cell types: osteoclasts (bone-resorbing cells), osteoblasts (bone-forming cells) and osteocytes. Modification of bone begins early in foetal life, and when the skeleton is fully developed in adults, almost all of the metabolic and mechanical activity has been nearly finished. The extracellular matrix of bone is constantly destroyed or rebuilt by osteoclasts and then replaced by osteoblasts. Osteoporosis, also known as bone loss, is a typical bone disease and reflects an imbalance in skeletal recirculation. The study of osteoblast differentiation from the earliest stages of differentiation can be carried out using embryonic stem cells (ESCs) isolated from mouse embryos. Those cells are pluripotent, which means they have the capacity to give rise to all types of cells of the body. Reports implicating Wnt signalling in the regulation of CatnB have prompted widespread interest in the use of Wnt molecules for the treatment of skeletal disorders. Wnt proteins comprise a large class of signalling molecules that regulate a variety of developmental processes. It is known that these Wnt proteins such as Wnt3a/Wnt5a are involved in the control of self-renewal of mesenchymal precursor cells, their early differentiation and lineage-specification into osteoblasts [1]. Thus, influencing the Wnt canonical and non-canonical signalling pathways could very well control the osteogenic differentiation process in vitro. Previous studies have shown that the canonical Wnt3a and the non-canonical Wnt5a are involved in the differentiation of murine embryonic stem cells (mESCs) in a differentiation stage-specific manner in the medium containing serum [2]. However, it is not clear whether these Wnts influence the differentiation process in a similar manner when using defined serum-free conditions. In the present study, I used recombinant Wnt3a and Wnt5a proteins and other factors, respectively, which influence Wnt signalling, together with Knockout Serum Replacer (KSR) as a combined treatment to induce early differentiation of the ESCs. These conditions were chosen to explore the effects of Wnts and serum on the process of differentiation. Taken together, our study showed that Wnt3a/5a together with serum replacer both enhance the level of osteoblast differentiation.

Der Knochen ist ein mineralisiertes Gewebe, das vielfache mechanische und metabolische Funktionen im Skelett ermöglicht. Das Skelettsystem enthält verschiedene Zelltypen: Osteoblasten (knochenbildende Zellen), Osteozyten (entstehen aus Osteoblasten und liegen in der mineralisierten Knochenmatrix vor), und Osteoklasten (Zellen der Knochensubstanz-Resorption). Die Entwicklung des Knochens beginnt früh im fötalen Stadium, wird ständig durch Osteoklasten zerstört und durch Osteoblasten wieder ersetzt. Viele Krankheiten der Knochensystems, z.B. die Osteoporose, auch bekannt als Knochenverlust, widerspiegeln eine Unausgewogenheit im Skelettumsatz. Für die Untersuchung von Osteoblasten der frühesten Entwicklungsstufen werden embryonale Stammzellen verwendet, welche pluripotent sind und die Kapazität besitzen, alle Arten von Zellen des Körpers zu bilden. Berichte schließen Wnt-Proteine in der Regulierung von ß-catenin Aktivierung ein und haben deswegen ein weit verbreitetes Interesse am Gebrauch von Wnt für die Behandlung von Knochenfunktionsstörung veranlasst. Wnt Proteine umfassen eine große Anzahl von Signalmolekülen, die eine Vielfalt von Entwicklungsprozessen regeln. Es ist bekannt, dass diese Wnt Proteine wie Wnt3a/Wnt5a an der Kontrolle der Selbsterneuerung von mesenchymalen Vorgänger-Zellen in der frühen Differenzierung und Abstammung von Osteoblasten beteiligt sind. So könnte der kanonische und nichtkanonische Signalweg den osteogenischen Differenzierungsprozess in vitro kontrollieren und beeinflussen [1]. Frühere Studien haben gezeigt, dass der kanonische Wnt3a und der nichtkanonische Wnt5a Signalweg an der Differenzierung von embryonalen Stammzellen in einer ganz spezifischen Weise im Serum beteiligt sind [2]. Jedoch ist es nicht bestätigt, ob diese Wnts die Differenzierung der embryonalen Stammzellen auf eine ähnliche Weise beeinflussen, wie das Verwenden von Medium ohne Serum. In der vorliegenden Studie verwendeten wir rekombinante Wnt3a und Wnt5a Proteine und andere Faktoren, welcher den Wnt-Signalweg zusammen mit dem Knock-Out-Serum-Ersatz entweder getrennt oder als eine vereinigte Behandlung beeinflussen, um eine frühe Differenzierung der Zellen zu veranlassen. Beide Bedingungen wurden gewählt, um die Effekte von Wnts und Serum auf dem Prozess der Differenzierung zu erforschen. Unsere Studie zeigte, dass Wnt3a/5a zusammen mit dem Serum-Ersatz das Niveau der Osteoblasten Differenzierung erhöhen.
