Characterization of IL-13- and allergen-induced airway hyperreactivity in precision-cut lung slices of different species
Abstract
Allergic asthma is a chronic inflammatory disease of the airways and is characterized by reversible airway obstruction, airway hyperresponsiveness (AHR) and airway inflammation. Distinctive features of asthma are eosinophilic invasion of the lung, increased mucus production and airway narrowing to non-specific stimuli. The outline of the airway inflammation during asthma includes several phases: induction of the allergic reaction, manifestation of the disease with early allergic reaction and late phase reaction. Several cell types and a broad range of cytokines are involved in asthma pathogenesis. Of special interest in allergic asthma is interleukin-13 (IL-13). IL-13 influences respiratory epithelial cells and airway smooth muscle cells leading to characteristic hallmarks in the airways of asthma patients, which can result in airway remodelling. The mechanism of asthma is not fully understood. To discern the underlying immunological mechanisms, suitable preclinical models to study asthma are required. One pre-clinical model to study airway diseases is the ex vivo model of the precision-cut lung slices (PCLS). In this master thesis different features of allergic asthma were mimicked in PCLS of rat or human lung tissue for pre-clinical drug development. Cells addressed in experiments of this master thesis are airway epithelial cells and smooth muscle cells. These cells were immunostained in human PCLS. Immediate allergic reaction in passively sensitized rat PCLS was established in slices of two rat strains with two different serums from ovalbumin-allergic rats. Bronchoconstriction in passively sensitized rat PCLS was inhibited with rat anti-Immunoglobulin E (IgE), serotonin-antagonist ketanserin and histamine-antagonist cetirizine. Inhibitory effects of dimethyl sulfoxide (DMSO) was detected at a specific concentration of 0.001 % DMSO. Colocalization of mast cells with IgE and histamine was observed. Airway hyperreactivity was induced with IL-13 in human PCLS and stimulation with methacholine was used to induce bronchoconstriction. AHR in human PCLS occurred 3 h and 6 h after IL-13 pre-incubation. Characteristic asthma feature of increased mucus production was induced with IL-13 in rat PCLS and identified by PAS staining. Mucin5AC hypersecretion was induced by incubation of human bronchus slices with IL-13. Influx of eosinophils into lung tissue was tried to mimic with PCLS lysates in a transwell migration assay. Human eosinophils, isolated from blood, had to migrate to eotaxin-3-rich PCLS lysate. Eosinophil migration to eotaxin-3-rich lysates from human PCLS could not be established successfully. In conclusion, human PCLS and bronchus slices could be used to mimic certain asthma features so far. Passively sensitized rat PCLS are a suitable model to study early allergic reactions.

Asthma ist eine chronische Entzündung der Atemwege, characterisiert durch reversible Obstruktion, Atemwegshyperreaktivität (AHR) und Atemwegsentzündungen, welche zu Mucus- Überproduktion, Eosinophilie und Atemwegsverängung durch unspezifische Reize f¨uhren können. Eine Atemwegsentzündung zeigt sich zu Beginn durch die Induktion der allergischen Reaktion, einer Frühphasen-Reaktion, gefolgt von einer Spätphasen-Reaktion. An der Asthmapathogenese ist eine Vielzahl von Zytokinen und Immunzellen beteiligt. Das Zytokin Interleukin-13 (IL-13) ist im allergischen Asthma von besonderem Interesse. IL-13 beeinflusst das respiratorische Epithel und die glatte Atemwegsmuskulatur, wodurch charakteristische Asthmafaktoren wie Mukusproduktion und Eosinophilie beeinflusst werden, was zu einer Atemwegsremodellierung führen kann. Der Gesamtmechanismus des Asthmas ist nicht vollständig aufgeklärt. Um die molekularbiologischen Hintergründe des Asthmas zu erforschen, sind geeignete Versuchsmodelle notwendig. Ein verifiziertes präklinisches ex vivo Model sind die Precision-Cut Lung Slices (PCLS). Für diese Masterarbeit wurde die Methode der PCLS herangezogen, um in diesen Gewebeschnitten Eigenschaften des Asthmas zu imitieren, welche zur präklinischen Pharmazeutika-Entwicklung genutzt werden k¨onnen. In den durchgef¨uhrten Versuchen wurden vor allem die respiratorischen Epithelzellen und die Zellen der glatten Atemwegsmuskulatur angesprochen. Das respiratorische Epithel und die glatte Atemwegsmuskulatur wurde in humanen PCLS immunfluoreszent gef¨arbt. In PCLS aus Rattenlungen wurden mit Hilfe von passiver Sensibilisierung mit Serum von Ovalbuminallergischen Ratten allergische Fr¨uhphasen-Reaktionen erzeugt. Die Bronchokonstriktion in passiv sensibilisierten Ratten-PCLS wurde durch Ketanserin, anti-IgE und Cetirizin inhibiert. Auch Dimethylsulfoxid (DMSO) zeigte in einer definierten Konzentration von 0,001 % eine inhibierende Wirkung. Eine Kolokalisation von IgE, Histamin und Mastzellen wurde mit Hilfe von Immunfluoreszenz-F¨arbungen gezeigt. Die Atemwegshyperreagibilität wurde in humanen PCLS zu den Zeitpunkten 3 h und 6 h, nach Inkubation mit IL-13, erzeugt. Die Bronchokonstriktion wurde durch Zugabe von Methacholin induziert. Eine Mucus- Überproduktion konnte ebenfalls durch IL- 13 Inkubation in Ratten- und humanen Lungen-Schnitten induziert werden. Eine erhöhte Mucusproduktion in Ratten-PCLS wurde mit Hilfe der PAS- Färbung detektiert. Eine Mucin5ACÜberproduktion wurde in humanen Bronchusschnitten durch IL-13 Inkubation induziert. Die Eosinophil-Migration wurde mit Eotaxin-3-angereicherten PCLS-Lysaten als Stimulus in einem Transwell-Migrationsversuch durchgeführt. Hierzu wurden Eosinophile aus humanem Blut isoliert. Die Migration von Eosinophilen zu Eotaxin-3 aus humanen PCLS konnte in einem Transwell-Migrationsversuch nicht erfolgreich durchgef¨uhrt werden. Zusammenfassend ist festzustellen, dass spezifische Eigenschaften des Asthmas in humanen PCLS und Bronchus induzierbar sind. Es konnte gezeigt werden, dass passiv sensibilisierte Ratten-PCLS ein geeignetesModel darstellen, um allergische Frühphasen- Reaktionen zu untersuchen.
