Abstract
(A1) Ein Verfahren zur Kryokonservierung von biologischem Material, das biologische Zellen (1) mit Zellmembranen (2), Zellkernen (3), Zytoplasma (4) und Zellorganellen (5) enthaelt, umfasst die Schritte Praeparation der biologischen Zellen (1), derart, dass die Zellmembranen (2) unterbrochen werden, Dehydrierung der Zellkerne (3) und Einstellung von Konservierungsbedingungen, unter denen die Zellkerne (3) einen kryokonservierten Zustand aufweisen. Des Weiteren koennen die Zellmembranen (2), Zytoplasma (4), Zellorganellen (5) und/oder Teile von diesen der Dehydrierung und der Kryokonservierung unterzogen werden. Es werden auch die Verwendung von Alkoholen, Proteinen, Zucker, Elektrolyten, Polyanionen, Polykationen, Polymeren, Oelen oder Gelen als Kryoprotektiva und eine Vorrichtung zur Krykonservierung von Zellkernen (3) biologischer Zellen (1) beschrieben.

DETAILED DESCRIPTION - An INDEPENDENT CLAIM is included for a device, for cryopreservation of cell nuclei of biological cells, comprising a device for preparing the biological cells, which disrupts the cell membrane of the biological cells, a device for dehydrogenating the cell nuclei, and a preserving device for adjusting the conditions under which the cell nuclei is present in cryopreserved state. USE - The method is useful for cryopreservation of biological materials comprising cells with cell membranes, cell nuclei, cytoplasm and cell organelles. The device is useful for cryopreservation of cell nuclei of biological cells. Alcohol, protein, sugar, electrolyte, polyanion, polycation, polymer, oil or gel is useful as cryoprotectants (all claimed). ADVANTAGE - The improved cryopreserving method has high level of reliability and reproducibility. The method avoids the use of cryoprotectants that affect the cells and their vitality rate. The cryoprotectants used have limited penetration via the cell membrane.
